Enhancement of in vitro drug release by using polyethylene glycol 400 and PEG-40 hydrogenated castor oil in pellets made by extrusion/spheronisation

摘要: Abstract The in vitro release rate of hydrochlorothiazide from Avicel ® PH101 pellets was enhanced by the incorporation polyethylene glycol 400 (PEG 400) and PEG-40 hydrogenated castor oil (Cremophor RH40) into formulation. Pellets containing more than 20% (w/w) PEG released greater 70% during first 5 min whereas without only 45% after 90 min. X-ray diffraction patterns showed that completely dissolved present formulations. Reducing amount formulation decreased hydrochlorothiazide. use pellet increased (30% min) due to solubilisation drug Cremophor RH40. An increase RH40/hydrochlorothiazide ratio Storage under ambient conditions for 6 months did not alter their dissolution profiles. 21% RH40 a storage conditions.
