Assessing Therapeutic Potential of Magnetic Mesoporous Nanoassemblies for Chemo-Resistant Tumors.

摘要: Smart drug delivery system with strategic distribution is the future state-of-the-art treatment for any malignancy. To investigate therapeutic potential of such nanoparticle mediated system, we examined efficacy dual drug-loaded, pH and thermo liable lipid coated mesoporous iron oxide-based magnetic nanoassemblies (DOX:TXL-LMMNA) in mice bearing both sensitive (A2780(S)) resistant (A2780-CisR) ovarian cancer tumor xenografts. In presence an external AC field (ACMF), DOX:TXL-LMMNA particles disintegrate to release encapsulated due hyperthermic temperatures (41-45 oC). vivo bio study utilizing optical properties demonstrated minimum organ specific toxicity. Noninvasive bioluminescence imaging A2780(S) tumors administered DOX-TXL-LMMNA followed by application ACMF revealed 65% less luminescence signal 80% showed complete regression within eight days. A six months follow-up absence relapse 70% mice. Interestingly, A2780-CisR which did not respond alone (DOX:TXL) reduction volume after thermo-chemotherapy Cytotoxic effect was more pronounced cells than their counterpart. Thus these novel stimuli hold great promise therapy malignancies clinical applications.
