摘要: Abstract Electrophoresis of DNA has been performed traditionally in either an agarose or acrylamide gel matrix. Considerable effort directed to improved quality agaroses capable high resolution, but for small fragments, such as those from polymerase chain reaction (PCR) and post-PCR digests, still offers the highest resolution. Although gels can easily be prepared open-faced format gain conveniences horizontal electrophoresis, does not polymerize presence air usual configurations preparation lead electrophoresis vertical dimension. We describe here a very simple device method prepare manipulate polyacrylamide (H-PAGE). In addition, arrangement enables loading arrays wells. Since many procedures are undertaken standard 96-well microtiter plates, we have also designed which preserves exact configuration 8 × 12 array tracks following 71.6° diagonal between wells (MADGE, electrophoresis), using agarose. This eliminates almost all staff time taken setup, loading, recordkeeping resolution genotyping pattern recognition. The nature size allow direct stacking one tank, so that tank used typically analyze 30-60 samples readily 1000-2000 samples. would enable robotic loading. allows analysis charge, parameters inaccessible liquid-phase methods: thus, patterns, variable length repeats, haplotypes is possible, well adaptability typing point variations protocols create difference detectable by electrophoresis.
