6-OHDA injections into A8-A9 dopaminergic neurons modelling early stages of Parkinson's disease increase the harmaline-induced tremor in rats.

摘要: The aim of the present study was to examine influence a unilateral 6-hydroxydopamine (6-OHDA)-induced partial lesion both substantia nigra pars compacta (SNc, A9) and retrorubral field (RRF, A8) on tremor evoked by harmaline. 6-OHDA (8μg/2μl) injected unilaterally into region posterior part SNc RRF. Harmaline administered in dose 7.5mg/kg ip eighth day after operation forelimbs, head trunk measured. We found that increased intensity induced harmaline but did not its character. Stereological examination extent revealed losses dopaminergic (tyrosine hydroxylase-immunoreactive) neurons anterior (30%) (72%) SNc, as well RRF (72% average). Levels dopamine all metabolites, noradrenaline concentrations, were ipsilaterally moderately decreased caudate-putamen lesioned animals, however, DOPAC cerebellum increased. In caudate-putamen, ipsi/contra ratio level correlated negatively, while turnover positively with intensity. However, an inverse relationship found. Moreover, this symptom serotonin negatively 5-HIAA/serotonin contralateral side cerebellum. results seem indicate modulation serotonergic transmissions modelling early stages Parkinson's disease may triggered
