摘要: A non-competitive immunoassay based on micellar electrokinetic capillary chromatography (MECC) with laser-induced fluorescence (LIF) detection has been developed for the determination of alpha-fetoprotein (AFP). The anti-AFP antibody was labeled fluorescein isothiocyanate (FITC) and product used as a fluorescent tracer, then AFP mixed antibody. After incubation, immune AFP–antibody complex separated from free by MECC. parameters affecting separation such concentration sodium dodecyl sulfate (SDS), buffer pH voltage were investigated following conditions selected: 20 mM tetraborate containing 100 SDS at 9.50, kV voltage. limit this assay 0.1 ng/ml linear range spanning two orders magnitude. This method applied to determine in human serum.
