A rapid, quantitative determination of total and free cholesterol with anthrone reagent

摘要: Abstract A simple, rapid microcolorimetric determination for total and free 3β-hydroxy sterols in serum tissues has been described. Following extraction of saponification the esterified fraction by established procedures, sterol is precipitated 15 min at 45°C as digitonide using 10% aluminum chloride. The isolated purified twice recrystallizing from methanol-10% chloride (1 : 1), each step requiring 20 45°C. precipitate washed with acetone suspended glacial acetic acid, digitonin moiety determined a stable anthrone reagent. Multiple analyses concentrations low 15–20 μg are routinely an accuracy comparable to method Sperry Webb, greater sensitivity precision than obtained latter method.
