摘要: The developing brain is highly vulnerable to the adverse effects of chemicals, resulting in neurodevelopmental disorders humans. Currently, animal experiments rat are gold standard for developmental neurotoxicity (DNT) testing; however, these guideline studies insufficient terms use, time and costs bear issue species extrapolation. Therefore, necessity alternative methods that predict DNT chemicals faster, cheaper with a high predictivity humans internationally agreed on. In this respect, we developed an vitro model key event screening, which based on primary human neural progenitor cells grown as neurospheres. They able mimic basic processes early fetal development enable investigation differences between rodents corresponding cellular models. goal study was investigate what extent neurospheres were correctly potential well-characterized training set nine by investigating cell proliferation, migration neuronal differentiation parallel viability, compare chemical responses We demonstrate (1) correlating results existing vivo data, classified most compounds thus may serve valuable component modular testing strategy (2) differed their sensitivity reflecting toxicodynamic chemicals.
