Increased vascular eNOS and cystathionine-γ-lyase protein after 6 weeks oral administration of 3, 5, 7, 3′, 4′-pentamethoxyflavone to middle-aged male rats

摘要: Effects of treatment middle-aged male rats with 3, 5, 7, 3′, 4′-pentamethoxyflavone (PMF) on vascular and perivascular adipose tissue (PVAT) functions blood chemistry were investigated. Rats received PMF (22 mg/kg), orally or vehicle, twice a day for 6 weeks. The PMF-treated had lower serum glucose, higher HDL-C levels, but no change in other parameters. Thoracic aortic mesenteric rings treated produced maximal contraction to phenylephrine that was normalized by NG-nitro-L-arginine (L-NA) endothelial removal. aortic- the showed improved relaxation acetylcholine, not glyceryl trinitrate, eNOS protein. DL-propargylglycine (PAG) caused greater increase baseline tension ring low concentrations phenylephrine. PVAT lowered contractile response L-NA pretreated both groups, PAG effect. cystathionine-γ-lyase (CSE) protein thoracic rings, PVAT, shows increased expression after treatment. Overall, middle aged appeared production NO H2S from vessels upregulating CSE. also decreased fasting glucose toxicity liver kidney functions. Thus, is novel compound possible use as health product prevent and/or reduce development diabetes type II cardiovascular disease.
