Three-dimensional cultures of dissociated myenteric plexus as amodel for the development of the enteric nervous system

摘要: Failures in the development of enteric nervous system (ENS) can lead to severe dis-eases, such as Hirschsprung`s disease or any other dysganglionosis. Trying imitate de-velopment ENS, myenteric plexus, vitro might give us a better understanding underlying problems and chance develop new diagnostic even therapeutic ap-proaches. Myenteric plexus newborn rats was isolated by enzymatical digestion, dissociated cultivated either on glass coverslips, three-dimensional gel extracellular-matrix-components. Supplementing culture media with neurotrophins Glial-cell-line-derived-neurotrophic-factor (GDNF) brain-derived-neurotrophic-factor (BDNF), alone com-bination, differences growth behaviour individual culture. GDNF increased survival, well neurite outgrowth neurons, while BDNF had no obvious effect when administered alone. Only combination it in-creased branching patterns neurites, indicating higher degree differentiation. These phenomena were seen glass, cultures. The only difference was, that cultures grown gels formed secondary ganglia which similar what is more cases dysganglionosis, there specific reaggregation glass. “artificial hypoganglionosis” varied, depending factors supple-mented media. Cultivating environment seem be valuable model so investigate respec-tively lack certain for an intact working gastrointes-tinal innervation.
