摘要: Glial cell-line-derived neurotrophic factor (GDNF), a member of the transforming growth-factor- (TGF-) β-family, is an essential for development enteric nervous system (ENS) during embryogenesis. In present study, effects GDNF on postnatal ENS were investigated using cultures myenteric plexus from small intestine newborn albino rats different developmental phases (P1, P7, P14). Myenteric was dissociated and cultivated as mixed neurons glial cells. After seeding, kept 24 h or 7 days in serum-free medium containing various doses (1, 10, 100 ng/ml) GDNF. The effect evaluated parameters such cell size, neuronal survival, neurite elongation. While neither glial-cell nor size influenced by GDNF, there observable upon survival treated with displayed increased outgrowth. promoting dose- age-dependent, decreasing clearly early period. Already after h, P1 but not P14 cultures. long-term cultures, marked tendency to form aggregates dense fiber networks observed when These observations suggest that plays important role only pre-, also system.
