Suppression of growth of PHA-stimulated human lymphocytes by a novel steroid (RU 28 362) lacking the 21-OH group.

摘要: RU 28 362 is a novel synthetic steroid (lacking the 21-OH group) which reacts exclusively with glucocorticoid receptor. It therefore chemically different from other natural and glucocorticoids in common use also bind to lesser extent receptors for classes of steroids. Aspects immunosuppressive effect were studied by comparing its suppressive effects on PHA-stimulated growth human lymphocytes those hydrocortisone, betamethasone inactive analogue, 21-deoxyhydrocortisone. was more potent than either hydrocortisone or betamethasone, but all three had parallel dose-response curve suppression increase cell volume that occurs activation during first day culture. In studies time appearance density IL-2 3 days culture, somewhat inhibitory steeper betamethasone. much inhibiting [3H]-TdR incorporation 3-day PHA stimulated cultures. 21-deoxyhydrocortisone did not inhibit any aspect lymphocyte growth.
