摘要: Abstract A simple, rapid and accurate method for the simultaneous determination of four purine pyrimidine bases (cytosine, 5-methylcytosine, adenine N6-methyladenine) has been developed. The quantitative these was accomplished by ion chromatography (IC) with direct conductivity detection (CD) based on their ionization in acidic medium without chemical suppression. recovery cytosine, calf thymus DNA more than 98% (n = 3) relative standard deviation (RSD, n = 5) less 2.4%. In a single chromatographic run, could be separated determined 10 min. limits were found to 0.05 μg/mL 0.08 μg/mL 0.07 μg/mL adenine, N6-methyladenine. Linear ranges 0.2–95.1 μg/mL cytosine (r2 = 0.9996), 0.3–196.6 μg/mL 5-methylcytosine (r2 = 0.9994), 0.3–105.5 μg/mL (r2 = 0.9998), 0.3–159.1 μg/mL N6-methyladenine (r2 = 0.9999). With proposed method, successfully detected DNA. We also using RP-HPLC. Compared RP-HPLC, IC offers advantages such as high selectivity simple mobile phase.
