Physically crosslinked injectable hydrogels for long-term delivery of oncolytic adenoviruses for cancer treatment

摘要: A dual pH- and temperature-responsive physically crosslinked injectable hydrogel system was developed for efficient long-term delivery of oncolytic adenoviruses (Ads). Three different types hydrogels with chemical compositions properties were prepared. These good biocompatibility can be injected at pH 9.0 room temperature rapidly form a gel under body or tumor microenvironment conditions. Ads encapsulated in released gradually without burst release. Moreover, these provided protective environment maintained their bioactivity long period time. Compared to naked Ads, protected by showed strong cytotoxicity cancer cells even after 11 days. The Ad-loaded also exhibited enhanced antitumor therapeutic effects human xenograft models. Due outstanding properties, might have potential treatment.
