Dystrophic phenotype improvement in the diaphragm muscle of mdx mice by diacerhein

摘要: Chronic inflammation and oxidative stress are striking features of Duchenne muscular dystrophy disease. Diacerhein is an anthraquinone, which exhibits anti-inflammatory antioxidant properties. Based on their actions, the present study evaluated effects diacerhein against myonecrosis, inflammatory response in diaphragm muscle mdx mice compared these results to current treatment widely used DMD patients, with a main focus impact prednisone. The demonstrated that prevented damage indicated by decrease IgG uptake fibers, lower CK levels serum, reduction fibers central nuclei concomitant increase peripheral nuclei. It also had effect process, decreasing area, macrophage staining TNF-α IL-1β content. Regarding stress, was effective reducing ROS lipid peroxidation from mice. Compared prednisone treatment, our findings improved dystrophic phenotype similar glucocorticoid therapy. In this respect, work suggests has potential use as alternative drug dystrophinopathy recommends its antioxidants properties should be better understood.
