摘要: Abstract 1. A new column chromatographic procedure for the separation of different hemoglobin fractions using DEAE-cellulose as adsorbent is described. Except fetal elution rates components were found to be in accordance with differences electrophoretic points. The rates, measured by their volumes, are within certain limits independent total amount Hb chromatographed; greatly influenced initial pH values adsorbent. 2. Examples separations presented cases following Hb-abnormalities: AS, AC, AJ (Georgia), SS, SC, normal cord blood, blood samples from babies heterozygous Hb-S and Hb-C, sample abnormal Bart's Hb, Thalassemia minor major, Hb-E-thalassemia disease, persistent high Hb-F abnormality, abnormality combined Hb-S, Hb-B2 trait, trait associated thalassemia, “Lepore” a case an unknown Hb-component. 3. Since small amounts generally eluted Hb-A1 fraction, application alkali denaturation method this isolated fraction offered fairly accurate determination Hb-F. Applications thalassemia have been presented. 4. quantitative data obtained agreement results starch block electrophoresis carboxymethylcellulose chromatography. It was emphasized that particular use fractions: A2, B2, Lepore 5. heterogeneity hemoglobins adults, sickle cell anemia patients, observed chromatography closely related those methods CM_cellulose 6. great preparation large quantities pure Hb-fractions: examples Hb-A2 Hb-Lepore
