摘要: Doxorubicin (DOX) is the best anticancer agent that has ever been used, but acquired tumor resistance and dose limiting toxicity are major road blocks. Concomitant use of natural compounds a promising strategy to overcome this problem. Neferine, proven found in green embryos lotus seed. The study demonstrates neferine acts as an effective enhancer DOX-induced cell death A549 cells through ROS mediated apoptosis with MAPK activation inhibition NF-jB nuclear translocation. Cotreatment significantly enhanced intracellular DOX-accumulation. Neferine DOX combination also triggered oxidative stress Ca 2+ accumulation dissipation mitochondrial membrane potential addition significant loss cellular antioxidant pool. inhibitor effectively decreased cell-death induced by DOX. Pretreatment glutathione reversed combined regimen recovered Bcl2/ Bax ratio. Moreover, treatment increased viability DOX-treated cardio- myocytes indicating possible protective role towards cardiotoxicity. Taken together, our results suggest using could be helpful increase efficacy achieve synergism curbing toxicity.
