摘要: This study determines the validity of utilizing radioimmunoassay CuZn and Mn superoxide dismutase in rat for defining mechanism control over mammalian tissue concentrations. To accomplish this, were purified. The dimer had a specific activity 3600 units/mg protein subunit Mr 17,000. tetramer 3700 22,000. Both enzymes provided single discrete band on disc gel electrophoresis. purified utilized to develop sensitive (less than 2.5 ng/ml less 3.12 dismutase) reproducible immunoassays specificity which was confirmed by homogenate dilution column chromatography. Immunoassay these tissues permitted clarification existing data based assays demonstrated trend higher concentrations high mitochondrial content (with relative comparable man) low islets (providing an explanation their sensitivity free radical damage). represents first report dismutase, second successful purification apparent purity stability). support proposition that radioimmunoassays rats will provide useful system investigation mechanisms tissues.
