摘要: Summary In Plasmodium falciparum , the deadliest form of human malaria, nuclear periphery has drawn much attention due to its role as a sub-nuclear compartment involved in virulence gene expression. Recent data have implicated components envelope regulating expression several eukaryotes. Special been given nucleoporins that compose pore complex (NPC). However, very little is known about parasites. Here we characterize PfSec13, an unusual nucleoporin P. which shows unique structural similarities suggesting it fusion between Sec13 and Nup145C yeast. Using super resolution fluorescence microscopy (3D-SIM) vivo imaging, show dynamic localization PfSec13 during parasites9 intra-erythrocytic development corresponds with NPCs these dynamics are associated microtubules rather than F-actin. addition, does not co-localize heterochormatin markers HP1 H3K9me3, euchromatic location NPCs. The proteins indicate this Nup cellular processes. Indeed, ultrastructural chromatin immunoprecipitation analyses revealed that, addition NPCs, found nucleoplasm where chromatin. Finally, used peptide nucleic acids (PNA) downregulate essential for parasite proliferation erythrocytes.
