Down-regulation of hypoxia-inducible factor-1 suppresses malignant biological behavior of triple-negative breast cancer cells

摘要: This study is to investigate the effect and mechanism of reduced hypoxia-inducible factor (HIF)-1a expression on malignant behavior MDA-MB-231 cells. HIF-1α was interfered by siRNA. Western blot used detect protein HIF-1α, active fragments caspase 3 vimentin. Cell count, flow cytometry Hoechst staining were evaluate cell growth apoptosis. Matrigel invasion wound scratch assay performed measure ability migration. After cells transfected with HIF-1α-targeted siRNA, successfully interrupted retarded. Compared random siRNA group, in group facilitated apoptosis but had no cycle. In addition, treated expressed (17 12 kD) after serum starvation for 0 60 h. Caspase activity further confirmed above finding. Reduced impaired migration invasiveness a reduction vimentin CK18 protein. Inhibition synthesis or enhancement its degradation reversed phenotypes could probably be potential means treatment triple-negative breast cancer.
