GABA UPTAKE IN RAT CENTRAL NERVOUS SYSTEM: COMPARISON OF UPTAKE IN SLICES AND HOMOGENATES AND THE EFFECTS OF SOME INHIBITORS

摘要: — Fifty-two substances were tested as inhibitors of the uptake [3H]GABA in slices rat cerebral cortex. Among GABA analogues tested, only 2-fluoro, 3-hydroxy and 2-amino compounds had affinities for mechanism comparable to that GABA. was also potently inhibited by p-chloromercuriphenylsulphonate, N-ethylmaleimide, chlorpromazine haloperidol. No found act a competitive manner with respect examined homogenates cortex other regions CNS. There rapid into particles when homogenate samples incubated labelled amino acid; this similar kinetic properties inhibitor sensitivity observed intact tissue. Density gradient centrifugation experiments indicated responsible probably synaptosomes.
