Presynaptic mitochondrial morphology in monkey prefrontal cortex correlates with working memory and is improved with estrogen treatment

摘要: Humans and nonhuman primates are vulnerable to age- menopause-related decline in working memory, a cognitive function reliant on the energy-demanding recurrent excitation of neurons within Brodmann’s Area 46 dorsolateral prefrontal cortex (dlPFC). Here, we tested hypothesis that number morphology (straight, curved, or donut-shaped) mitochondria dlPFC presynaptic boutons altered with aging menopause rhesus monkeys (Macaca mulatta) these metrics correlate delayed response (DR) accuracy, well-characterized measure dlPFC-dependent memory. Although bouton density size was not significantly different across groups distinguished by age menses status, DR accuracy correlated positively total straight per bouton. In contrast, inversely frequency containing donut-shaped mitochondria, which exhibited smaller active zone areas fewer docked synaptic vesicles than those curved mitochondria. We then examined effects estrogen administration test whether treatment known improve memory influences mitochondrial morphology. Aged ovariectomized treated vehicle displayed significant impairment concomitant 44% increase both were reversed cyclic estradiol treatment. Together, our data suggest hormone replacement therapy may benefit aging, part promoting health dlPFC.
