Disruption of peroxisome function leads to metabolic stress, mTOR inhibition, and lethality in liver cancer cells

摘要: Peroxisome houses a large number of enzymes involved in lipid and phytochemical oxidation as well synthesis bile acid other specialized lipids. resident are imported into the organelle via conserved cargo transport system composed many peroxins, protein factors essential for biogenesis peroxisome. Among PEX5 plays transporter role, PEX2, 10, 12 thought to form complex that functions an E3 ubiquitin ligase help recycle modification-dependent process. Previous studies have demonstrated importance peroxins postnatal development especially nerve systems. These also show or function peroxisomes is dispensable cellular viability. In contrast, however, we report here PEX2 viability liver cancer cells, probably through altering metabolism signaling pathways. Our results suggest may be potential targets therapeutics against cancer.
