摘要: We explored the state of p53 gene in gastric cancer. Using one or more methods, we examined 15 specimens from primary carcinomas (14 tumors, cell line), five lines derived metastases, and seven paired samples nonmalignant mucosa. Sequence analyses complementary DNA containing entire open reading frame demonstrated abnormalities tumors all metastases. The single line a carcinoma had no abnormality gene. six included four point mutations, base-pair deletion resulting shift, 24 caused by an intronic mutation (as determined sequence analysis genomic DNA). Four mutations mapped to regions highly conserved among species involved simian virus 40 T-antigen binding. Restriction fragment length polymorphism studies confirmed that chromosome 17p allelic deletions occur only minority but they may frequently Northern blotting ribonuclease protection assays detected fraction sequencing. Our findings indicate are relatively rare appear be frequent metastatic lesions. results help understanding molecular events associated with progression metastasis carcinoma.
