摘要: The MET oncogene, encoding the tyrosine kinase receptor for hepatocyte growth factor/scatter factor, is expressed in epithelial cells and overexpressed a significant proportion of human cancers, suggesting occurrence transcriptional alteration(s). To identify promoter, we isolated recombinant cDNA clones encompassing entire 5'-noncoding sequence messenger RNAs. Using probes derived from this region, cloned genomic region spanning first exon flanking regulatory sequences. exon, containing untranslated sequence, present mRNAs 7.1, 5.9, 4.6 kilobases, showing that expression multiple transcripts regulated by single promoter. start site transcription was determined primer extension rapid amplification ends. We show 300-base pair fragment, sequences upstream site, efficiently drives reporter gene transfected cells. This promoter fragment also contains cis-acting elements responsible phorbol-ester induction.
