Inhibition of the Wnt/β-catenin pathway by the WWOX tumor suppressor protein

摘要: The WWOX gene encodes a candidate tumor suppressor protein (WWOX) implicated in variety of human diseases such as cancer. To better understand the molecular mechanisms action, we investigated novel partners this protein. Using two-hybrid system and coimmunoprecipitation assay, observed physical association between Dishevelled (Dvl) family signaling elements involved Wnt/β-catenin pathway. We found that enforced expression inhibited, inhibition endogenous stimulated transcriptional activity Inhibition also enhanced effect Wnt-3a on β-catenin stability. Moreover, sequestration Dvl-2 wild type Dvl-2NESm, mutated form predominantly localized nucleus, cytoplasm compartment by WWOX. Our results indicate is inhibitor would act, at least part, preventing nuclear import Dvl proteins.
