摘要: Circulating factor VIII (fVIII) levels increase during inflammation suggesting that fVIII synthesis or secretion is stimulated acute inflammation. To examine the mechanisms underlying this in circulating VIII, we have developed a sensitive and reliable semiquantitative assay for mRNA utilising competitive reverse transcriptase-polymerase chain reaction (RT-PCR, used to study two human liver cell lines, Hep-G2 Chang Liver cells. These cells were cultured under basal conditions following treatment with interleukin-1, -2 6 (IL-1, -2, -6). Following 18h culture IL-6 (maximum concentration 40 U/ml), these had risen 9 fold respectively, no concomitant rise control RNA levels. The dose responses both types similar, an ED50 of 11 U/ml. time course response was also similar lines first reaching significance by 3 h, maximum 12 h. IL-1 IL-2 effect at any doses studied. This provides evidence regulated expression suggests increased plasma phase may be due mRNA.
