摘要: Modulation of gap junctional intercellular communication (GJIC) is a known cellular event associated with tumor promotion. The present study was undertaken to test the potential preventive effect mushroom Phellinus linteus extract (PL) on inhibition GJIC, induced by hydrogen peroxide (H2O2), in WB-F344 rat liver epithelial cells (WB cells). Cells were pre-incubated PL (5 and 25 µg/ml) for 24 h this followed co-treatment H2O2 (500 µM) 1 h. (at 5 prevented GJIC blocked hyperphosphorylation connexin 43 H2O2. Moreover, activated p38 kinase, extracellular signal-regulated protein kinases (ERK)1/2 c-Jun N-terminal kinase (JNK) WB cells. indicates that able inactivate both ERK1/2 MAP kinases. However, did not affect JNK pathway. For reason, elucidate relation between we treated PD98059 (an MEK inhibitor) SB202190 (a inhibitor). These inhibitors also found prevent H2O2, which suggests may act as natural anticancer product preventing through inactivation In addition, our results indicate signaling pathway be closely related functionally junction
