Detection of diffusible ions in insect osmoregulatory systems by electron probe X-ray microanalysis using scanning electron microscopy and a cryoscopic technique

摘要: A technique for using electron probe X-ray microanalysis the detection of diffusible ions in frozen bulk specimens is described. The method has been applied to study insect osmoregulatory systems and preliminary results are presented Ephydrella larvae (Ephydridae, Diptera) which normally live salt water, Tenebrio (Tenebrionidae, Coleoptera). In larva, differences demonstrated (a) ionic composition different regions gut, (b) concentration Na+ Cl− hindgut lumen compared with blood, (c) cell types (d) concentrations cells correlated environmental salinity. larvae, high levels K+ region leptophragmata Malpighian tubules lends support hypothesis that these recruit from haemolymph.
