摘要: Primordial germ cells (PGCs) are progenitors of the cell lineage, giving rise to either spermatogonia or oogonia after completion gonadal differentiation. Currently, there is little information on mechanism PGCs migration leading formation primordial gonad in perciform fish. Yellowtail kingfish (Seriola lalandi) (YTK) (order Perciforms) inhabit tropical and temperate waters southern hemisphere. Fundamental details into molecular basis larval development this species can be easily studied Australia, as they commercially cultured readily available. In study, histological analysis YTK larvae revealed critical time points for genital ridge, resulting subsequent gonad. at 3, 5, 7 10 days post hatch (DPH), were not yet enclosed by somatic cells, indicating had started form. While 15, 18 20 DPH already settled ridge become develop. A higher number observed 15 proliferation, which corresponds with them becoming cells. Directional toward a event zebrafish, mouse chicken, stromal-cell derived factor (SDF1) signalling one key molecules PGC migration. We subsequently isolated from SDF1 (Slal-SDF1) gene, encodes 98-residue precursor protein signal peptide N-terminus. There spatial conservation between fish four cysteine residues positions C9, C11, C34 C49, expected form disulphide bonds stabilize SDF structure. YTK, Slal-SDF1 gene expression analyses shows that expressed 1 22 demonstrates distinct localisation DPH. These results provide platform further studies machinery yellowtail kingfish, well other species.
