摘要: An impressive body of evidence has been accumulated indicating that local protein synthesis is implicated in navigation neurite extension induced by guidance cues, such as semaphorin3A (Sema3A). We found previously a Src type tyrosine kinase Fyn and cyclin-dependent 5 (Cdk5) mediate Sema3A-signaling. also showed Sema3A elicits axonal transport through neuropilin-1, receptor for Sema3A, located at the growth cones. Here, we investigate relationship between Sema3A-induced signaling, synthesis, transport. Lavendustin A, inhibitor, olomoucine, suppressed facilitation anterograde retrograde dorsal root ganglion (DRG) neuron with without cell body. was attenuated DRG neurons fyn- (fyn-/-) Cdk5 activator, p35 (p35-/-)-deficient mice when compared those wild-type or heterozygous mice. Inhibition enhanced level immunoreactivity phosphorylated eukaryotic translation initiation factor 4E (eIF-4E) within min cones time course similar to facilitated This signal phospho-eIF4E blocked lavendustin A olomoucine not detected fyn-/- p35-/- neurons.
