Optimization of preparation of DHAQ-loaded PEG-PLGA-PEG nonaparticles using central composite design

摘要: Mitoxantrone (DHAQ)-loaded poly (ethylene glycol)-poly (lactic acid-co-glycolic acid) –poly (ethyleneglycol) (PELGE) nanoparticles (NP) were fabricated using an emulsification/solvent evaporation technique. A central composite design (CCD) was applied to evaluate the joint influence of three formulation variables: amounts polymer, concentration DHAQ, and ratio organic phase (inner-phase) aqueous (outer-phase). In this study, we optimize preparation technology on basis single factor evaluation. The optimal conditions for DHAQ-loaded nanoparticle found be: PELGE 9 mg/mL, inner-phase DHAQ 27.5 mg/L, inner-phase/outer-phase 8.5/1. results showed that CCD is ideal technique studies. entrapment efficiency (ER) 90% particle sizes are less than 500 nm. nanoparticles, as examined by transmission electron microscopy (TEM), have a smooth spherical surface. could be loaded into copolymers. nanoparticle-polymer delivery system established polymers carrier material.
