Helicobacter pylori antigen HP0986 (TieA) interacts with cultured gastric epithelial cells and induces IL8 secretion via NF-κB mediated pathway.

摘要: Background The envisaged roles and partly understood functional properties of Helicobacter pylori protein HP0986 are significant in the context proinflammatory or proapoptotic activities, two important facilitators pathogen survival persistence. In addition, sequence analysis this gene predicts a restriction endonuclease function which remained unknown thus far. To evaluate role gastric inflammation, we studied its expression profile using large number clinical isolates but limited biopsies patient sera. Also, antigenic altering cytokine responses human epithelial (AGS) cells including interaction with localization within AGS cells. Materials Methods For vitro study HP0986, 110 H. pylori were cultured from patients dyspepsia. For by qRT PCR 10 biopsy specimens studied. was also used to detect antibodies incubated recombinant determine response NF-κB activation. Transient transfection cloned pEGFPN1 subcellular homing cells. Results Out strains, 34 (31%) positive for observation correlated profiles. mRNA detected 7 specimens. Further, induced IL-8 secretion dose time-dependent manner via pathway. Serum against positively associated patients. revealed both cytoplasmic nuclear HP0986. Conclusion HP0986 moderately prevalent cultures points being naturally expressed. Collective observations induction TNFR1 NF-κB, localization, seropositivity data point gastroduodenal inflammation. We propose name gene/protein as ‘TNFR1 interacting A (TieA tieA)’.
