Quantitative methylation analysis of resection margins and lymph nodes in oral squamous cell carcinoma

摘要: Resection is the preferred treatment for oral squamous cell carcinoma, and pathological staging of resected specimen crucial. The role molecular biology in diagnosis minimal residual disease has not been fully investigated may improve staging. Multiple adjacent specimens were taken from tumour, invasive front, surgical margin, lymph nodes 20 patients with cancer. Bisulphite-treated DNA these was assayed quantitatively pyrosequencing methylation assays (PMA) CpG islands within gene promoters p16 CYGB genes. Results recorded histopathological results, compared clinical outcome. Biological technical replicates confirmed reliability techniques. PMA upgraded 13 margins, 6 which subsequently had a recurrent tumour. Not all recurrences predicted effects adjuvant make firm conclusions difficult.
