摘要: The regulation of the Na+-dependent glutamate/aspartate transporter system GLAST expressed in rat and mouse cerebellar cortical astrocytic cultures was examined. Pretreatment cells with l-glutamate 12-O-tetradecanoyl-phorbol-13-acetate (TPA), a known Ca2+/ diacylglicerol-dependent protein kinase (PKC) activator, produced decrease [3H]-d-aspartate uptake. This reduction dose- time-dependent sensitive to PKC inhibitors. Furthermore, l-glutamate–dependent uptake is non-receptor dependent process, because neither agonists or antagonists were effective mimicking reverting effect. Interestingly, transportable substrates could reproduce In sharp contrast, astrocytes, both TPA pre-exposure result an augmentation These findings suggest that glutamate undergoes region-specific regulation.
