High-dose immunosuppressant alters the immunological status of New Zealand white rabbits following skin transplantation

摘要: The aim of this study was to investigate the effect an immunosuppressant on immunological status New Zealand white rabbits after skin grafting, and evaluate a method for monitoring subjects with transplants. were randomly divided into allograft rejection, autograft tolerance, nontransplant, low-dose high-dose groups. in low- groups treated cyclosporine A intravenously 8 h prior transplantation once daily following at doses 2 25 mg/kg, respectively. At 12 days transplantation, spleens donor (female) recipient (male) harvested preparation single-cell suspensions. splenocytes from labeled 0.3 or 6 µM carboxy fluorescein diacetate succinimidyl ester, respectively, mixed cell suspension prepared. final injected rabbits. ratio two fluorescently populations peripheral blood measured using flow cytometry 1, 2, 4 injection, death rate calculated. Histological analysis also performed samples collected time splenectomy. rates rejection reached their highest levels injection spleen suspension. Allogeneic cells male almost completely removed within injection. increased slowly without specificity. This provides specific vivo patients grafting. can quickly accurately detect recipients splenocyte suspension, thereby indicating strength immune by systems recipients.
