摘要: As genetic erosion of pistachio (Pistacia vera L.) has been occurring in the Mediterranean, Central and West Asia North Africa, experiments were conducted to conserve two cultivars (‘Atli’ ‘Siirt’) mature germplasm by assessing both medium- long-term conservation techniques. In medium-term conservation, our results showed that it was feasible form either microshoots or encapsulated shoot apices up 12 months at 4°C dark. regards encapsulation-dehydration droplet-vitrification techniques assessed for cryopreservation cold-hardened osmoprotected ‘Atli’ cultivar. Among methods tested, 13.6% regrowth achieved with incubation explants droplets vitrification solution 150 min 0°C followed direct immersion liquid nitrogen (LN), rapidly thawed then cultured on Murashige Skoog’s (MS) medium containing 1 mg L−1 BA 0.5 mg L−1 GA3. The developed technique appeared as a promising procedure preservation germplasm. Moreover, assesment fidelity Random Amplified Polymorphic DNA analysis (RAPD) revealed out high levels stability between donor plant cryopreserved plants (similarity indexes 0.959 0.973) after they subcultured least 3 months. detected low level instability could be due toxic effect PVS2 regeneration phase. optimized techniques, especially slow growth storage, applied preserve other Pistacia species.
