摘要: The yeast Yarrowia lipolytica accumulates oils and is able to produce extracellular lipases when growing in different carbon sources including glycerol, the principal by-product of biodiesel industry. In this study, biomass production a novel mutant strain Y. was statistically optimized by Response Surface Methodology media containing biodiesel-derived glycerol as main source. This exhibited distinctive morphological fatty acid profile characteristics, showed an increased lipase activity. An organic source nitrogen addition 1.0 g/l olive oil were necessary for significant production. Plackett-Burman Central Composite Statistical Designs employed screening optimization fermentation shaken flasks cultures, maximum values obtained 16.1 12.2 Units/ml lipase, respectively. Optimized batch bioprocess thereafter scaled aerated bioreactors reached specific activity after 95 % had been consumed, three-fold higher than those cultures.
