摘要: Histone posttranslational modifications (PTMs) play a pivotal role in regulating the dynamics and function of chromatin. Supported by an increasing body literature, histone PTMs such as methylation acetylation together context “histone code,” which is read, or interpreted, effector proteins that then drive functional output chromatin (e.g., gene transcription). A growing number domains interact with histones and/or their have been identified. While significant advances made our understanding how these histones, wide putative histone-binding motifs yet to be characterized, undoubtedly, novel will continue discovered. In this chapter, we provide detailed method for construction combinatorially modified peptides, microarray fabrication using methods characterize interaction proteins, antibodies, substrate specificity histone-modifying enzymes. We discuss other available technologies user-friendly approach enable exploration histone–protein–enzyme interactions function.
