摘要: We describe a method for the specific isolation of representative N-terminal peptides proteins and their proteolytic fragments. Their is based on gel-free, peptide-centric proteomics approach using principle diagonal chromatography. will indicate that introduction an altered chemical property to internal holding free alpha-N-terminus results in column retention these peptides, thereby enabling further characterization by mass spectrometry peptides. Besides pointing changes protein expression levels when performing such proteome surveys differential modus, protease specificity substrate repertoires can be allocated since both are specified neo-N-termini generated after cleavage event. As such, our gel-free technology widely applicable amenable variety proteome-driven degradomics research.
