1,25-dihydroxyvitamin D3 specifically binds to a human breast cancer cell line (T47D) and stimulates growth

摘要: Abstract The T47D human breast cancer cell line contains a specific binding protein for 1.25-(OH) 2 D 3 , with 15000 sites per cell. Kd (1.1 × 10 −10 M) and sedimentation coefficient on sucrose gradients (3.7S) are the same as those reported 1,25-(OH) receptor in other tissues. Other vitamin metabolites bound to an order of affinities > 1,24,25-(OH) 25-OHD 24,25-(OH) . A new analogue 1β,25-(OH) was only effective at displacing hormone from receptor. Cell growth stimulated dose dependent manner by addition (up 0.8 nM) medium. higher concentration without effect.
