摘要: Publisher Summary The Semliki forest virus (SFV) is a positive-stranded RNA belonging to the alphaviruses. SFV expression system based on cDNA copy of viral genome. has been cloned into an SP6-based transcription vector in such way that exact copies alphavirus genome can be transcribed vitro. recombinant plasmid then serves as template for vitro synthesis RNA. When introduced cells, self-replicates, it codes its own replicase, leading high heterologous protein while competing out host synthesis. In SFVC vector, capsid gene retained and hierologist directly downstream by polymerase chain reaction (PCR). One needs avoid repeated freezing thawing stock. This will reduce infectivity. For use electroporation, cells should not passaged more than about 30 times.
