摘要: Publisher Summary Amino acid analysis on polyvinylidene difluoride (PVDF) has been performed with high protein concentrations, but the accurate analyses of lower amounts have more problematic. This chapter describes a reproducible manual hydrolysis method for determining amino composition PVDF bound one microgram. Five standard proteins ranging in molecular weight from 14.3 kD to 77 were electroblotted onto SDS PAGE and then hydrolyzed, quantitatively eluted analyzed by PTC analysis. The accuracy, precision, recovery at 0.5, 1.0 2.0 micrograms determined. An 1 μg can be determined 85% average 5% 88% recovery. Lower levels (0.5 jug) approximately 79% 11% 32%
