摘要: The ability to detect and quantify proteins of individual cells in high throughput is enormous biological clinical relevance. Most methods currently use either require the measurement large cell populations or are limited investigation few at a time. In this report, we present combination polydimethylsiloxane-based microfluidic device matrix-assisted laser desorption ionization time-of-flight mass spectrometer (MALDI-TOF-MS) that allows detection as 300 molecules peptide level ∼106 107 protein level. Moreover, performed an immunoassay with subsequent MALDI-TOF-MS capture insulin immobilized on surface (∼0.05 mm2) limit 106 molecules. This microfluidic-based approach therefore begins sample handling sensitivity requirements for MS-based single-cell analysis peptides holds potential easy parallelization immunoassays other highly sensitive analyses.
