摘要: Recently, mice and embryonic stem (ES) cells with allelic polymorphisms have been used extensively in the field of genetics developmental biology. In this study, we examined whether intersubspecific hybrid ES these genotypes can be efficiently produced by intracytoplasmic sperm injection (ICSI). Frozen-thawed spermatozoa from wild-derived strains, JF1 (Mus musculus molossinus), MSM (M. m. HMI castaneus), SWN spp.), were directly injected into mature oocytes laboratory ((C57BL/6 DBA2)F1; M. domesticus). The vitro vivo capacity F1 embryos was not significantly different among groups (P >0.05), term offspring obtained all (27%-34% transferred embryos). However, mean body placental weights differed genotype <5 10 ), greatest both weights. an application study using offspring, analyzed their mitochondrial DNA found consistent disappearance progeny. a second series experi- ments, generated blastocysts injecting C57BL/6 them to generate cell lines. lines esta- blished at high efficiency (9 20 blastocysts) confirmed. Thus, ICSI cryopreserved allows efficient immediate production number lines, which for polymorphic analysis mouse genetics. Mol. Reprod. Dev. 74: 1081-1088, 2007. 2007 Wiley-Liss, Inc.
