摘要: The utilization of small interfering RNAs (siRNAs) represents a new paradigm in gene knockout technology. siRNAs can be used to knockdown the expression particular by targeting mRNA post-transcriptional manner. While there are plethora reports applying siRNA-mediated silencing (PTGS) therapeutically apparent limitations such as duration effect and saturation RNA-induced complex (RISC). Recently, data have emerged that indicate an alternative pathway is operative human cells where been shown, similar plants, Drosophila, C. elegans, S. Pombe, mediate transcriptional (TGS). TGS antisense strand siRNA chromatin remodeling complexes specific promoter region(s). This results epigenetic modifications lead rewriting local histone code, silent state marks, ultimately heterochromatization targeted gene. observation siRNA-directed via suggests genes may also able silenced more permanently or for longer periods following single treatment fact offer therapeutic avenue could prove robust immeasurable value directed control target expression.
