摘要: In addition to membrane immunoglobulin (mIg), the B-cell antigen receptor contains Ig-alpha/Ig-beta heterodimers that link mIg intracellular signaling molecules. To compare ability of cytoplasmic domains Ig-alpha and Ig-beta transduce signals in B- T-cells, we constructed chimeric genes encoding extracellular transmembrane human CD8 alpha domain murine (CD8/Ig-alpha) or (CD8/Ig-beta). hybridoma LK 35.2 cells, antibody-mediated cross-linking mIg, CD8/Ig-alpha, CD8/Ig-beta induced similar increases calcium levels protein tyrosine phosphorylation. Substitution alanine for conserved leucine, but not isoleucine, residue within putative activation motif destroyed ability. T-cell DO-11.10 receptor, stimulated equivalent phosphorylation interleukin-2 production. Thus, are equally capable initiating early events downstream receptors as well evoking a complete biological effector response lymphocytes.
