Characteristics of an amidase isolated from a soil bacterium

摘要: Abstract Amidase was extracted from a bacterium isolated soil, and its properties were compared with those of amidase in soil. activity the bacterial protein lower than that soil amidase, respectively, optimal pH (7.0 vs 8.5), temperature (50 60°C), K m constant calculated by Lineweaver-Burk plot (5.6 17.9 mm), activation energy (18.9 43.3 kJmol − ) Q 10 (av. = 1.28 1.75). Bacterial stable at temperatures ranging to 50°C denatured 55°C. Toluene inhibited both amidase. When inhibitions 21 trace elements using 2 μmol 0.1 mg −1 protein, most effective inhibitors (> 25% inhibition) were: Ag(I), Cd(II), Cu(II), Hg(II), Ni(II), Pb(II), Zn(II), Al(III) Se(IV). The effect 16 pesticides on varied considerably. By μg active ingredient pesticide inhibition ranged 7 49% Dinitroamine Butylate, respectively. results show constituents have considerable influence reaction catalyzed this enzyme.
