摘要: Neuroblastoma cells are widely utilized models for the study of neuritic outgrowth phase neuronal differentiation, but relatively few such studies have attempted to identify nature process outgrowths. This identification will be necessary in developing strategies utilizing these distinguish underlying mechanisms involved axonogenesis vs dendritogenesis. In an effort procedures inducing specific types neurite outgrowth, and distinguishing axon- from dendrite-like processes, we subjected two neuroblastoma cell lines a variety stimuli previously shown induce cells. These include neuraminidase, ionomycin, KCl+dibutyryl cAMP, cholera toxin B subunit, retinoic acid, dibutyryl cAMP (alone), GM1 ganglioside, low serum. The first four (group 1) gave rise neurites with axon-like characteristics, including immunostaining that was positive phosphorylated high molecular weight neurofilament protein (NF-H) synaptic vesicle protein-2 (SV2), negative microtubule-associated (MAP2). next three treatments 2) resulted as evidenced MAP2 NF-H SV2. Neurites produced by serum had mixed properties. cytoskeletal differences were supported immunoblot analysis antisera above proteins. Striking morphological also noted, group 2-induced being significantly shorter more branch points than those generated 1 stimulants. Dendrite-like on other hand, does appear depend altered Ca2+.
