Identification of LEA, a podocalyxin‐like glycoprotein, as a predictor for the progression of colorectal cancer

摘要: Large external antigen (LEA) is considered as a colorectal cancer (CRC)-associated antigen, which was found via mAb ND-1 generated using hybridoma technology, but its molecular features remain unknown. To facilitate the clinical application of LEA, we identified LEA podocalyxin-like protein 1 (PODXL) with weight approximately 230 kDa, hyperglycosylated protein, immunoprecipitation and mass spectrometry in combination, verified that ND-1-recognized epitope on terminal sialic acid LEA. Correlation analysis between PODXL weight, immunological cross-reactivity, gene expression dependence supported identity Moreover, assessed significance 89 pairs primary CRC tissues adjacent nontumor by quantum dot-based immunohistochemistry (QD-IHC). High correlated significantly T stage (P = 0.010). Patients high showed poorer prognosis than those low 0.007). Multivariate indicated an independent predictor. Furthermore, comparative ND-1-based IHC toward sugar residue has higher sensitivity specificity to evaluate LEA/PODXL 3D3-based method core cell lines samples. In addition, first can be secreted exosomes from cells patient peripheral blood. Our results demonstrate predictor for progression potential applied setting sensitivity, specificity, noninvasive access.
