Morphine-induced conditioned place preference in mice: Metabolomic profiling of brain tissue to find “molecular switch” of drug abuse by gas chromatography/mass spectrometry

摘要: Conditioned place preference (CPP) is a widely used model to explore the mechanism of context-dependent learning. In this work, we developed GC-MS method investigate metabolites in mice brain which was study learning associated with rewarding effect morphine. Metabolites were extracted from tissues and derivatized followed by analysis gas chromatography/mass spectrometry (GC-MS). total, 69 peaks identified as known compounds. By Wilcoxon ran sum test p value ≤0.05, 21 selected considered potential biomarkers morphine brain. Using principal component (PCA) receiver-operator characteristic (ROC) curves, constructed combination these metabolic markers. Multivariate statistics yielded separation between two groups an area under curve 0.947. Some further discussed detail about their pathway. Results showed that our technique can be successfully applied profile for understanding molecular mechanisms drug abuse.
